Neurobiology of Disease Calcineurin in Reactive Astrocytes Plays a Key Role in the Interplay between Proinflammatory and Anti-Inflammatory Signals

چکیده
Maladaptive inflammation is a major suspect in progressive neurodegeneration, but the underlying mechanisms are difficult to envisage in part because reactive glial cells at lesion sites secrete both proinflammatory and anti-inflammatory mediators. We now report that astrocytes modulate neuronal resilience to inflammatory insults through the phosphatase calcineurin. In quiescent astrocytes, inflammatory mediators such as tumor necrosis factor(TNF) recruits calcineurin to stimulate a canonical inflammatory pathway involving the transcription factors nuclear factor B (NF B) and nuclear factor of activated T-cells (NFAT). However, in reactive astrocytes, local anti-inflammatory mediators such as insulin-like growth factor I also recruit calcineurin but, in this case, to inhibit NF B/NFAT. Proof of concept experiments in vitro showed that expression of constitutively active calcineurin in astrocytes abrogated the inflammatory response after TNFor endotoxins and markedly enhanced neuronal survival. Furthermore, regulated expression of constitutively active calcineurin in astrocytes markedly reduced inflammatory injury in transgenic mice, in a calcineurin-dependent manner. These results suggest that calcineurin forms part of a molecular pathway whereby reactive astrocytes determine the outcome of the neuroinflammatory process by directing it toward either its resolution or its progression.

Calcineurin (CaN) is a Ca2+/calmodulin (Ca2+/CaM)-dependent serine/threonine protein phosphatase expressed in most mammalian tissues but found at higher concentration in brain. In the last decade there have been a steadily increasing number of studies identifying neuronal CaN as a primary suspect in neuronal vulnerability, synapse loss, dendritic atrophy, synaptic dysfunction and neuroinflammat...
متن کاملRole of Propolis as a pharmaceutical candidate on interleukin-1β proinflammatory cytokine expression in Alzheimer's rat
Background: One of the important factors that play a key role in the pathogenesis of Alzheimer's disease (AD) is inflammatory processes leading to impaired expression of inflammatory cytokines such as interleukin-6, TNF-α, and interleukin-1β (IL-1β). Propolis, as a therapeutic compound, has anti-inflammatory and antioxidant properties. Therefore, the aim of the present study was to investigate ...

Thrombin is a multifunctional enzyme which has key roles in coagulation cascade and inflammatory events. The pro-inflammatory functions of thrombin occur by different mechanisms including increasing mast cell degranulation, up-regulating the expression of cell adhesion molecules (CAMs) and promoting the secretion of inflammatory chemokines and cytokines. Dysregulated signaling functions of thro...
